Одна из основных характеристик данной молекулы ДНК (или гибридного ДНК/РНК-дуплекса) - температура, при которой происходит диссоциация 50% двойной спирали, специфична для ДНК данного вида организмов, т.к. зависит от нуклеотидного состава и ее общих размеров; Тm отражает АТ/ГЦ-соотношение (AT/GC ratio) в молекуле нуклеиновой кислоты, т.к. пара Г-Ц имеет 3 водородные связи (А-Т - 2) и взаимодействие между нуклеотидами этой пары более сильное, - соответственно, Т.п. положительно коррелирует с долей пары Г-Ц в молекуле ДНК.* * *Тm — температура, при которой популяция двунитчатых молекул нуклеиновых кислот превращается в популяцию, наполовину состоящую из однонитчатых молекул; обозначает температуру плавления для такой системы.Температура плавления, Тпл — температура, при которой 50% имеющихся дуплексных ДНК или гибридных ДНК/РНК молекул диссоциируют (распадаются, разъединяются) на одиночные нити. Для определения Тпл раствор ДНК нагревается и при 260 нм измеряется ее спектр поглощения. Переход из двунитчатой в однонитчатую ДНК происходит в очень узком интервале температур и проявляется увеличением поглощения при 260 нм. При этом образуется сигмоидальная кривая (S-образная). Тпл определяется по средней точке возросшего поглощения тепла, что соответствует температуре, при которой 50% молекул диссоциирует. Для ДНК каждого вида организмов характерна своя Тпл, зависящая от состава оснований и ее общих размеров. Тпл отражает соотношение AT/GC в молекуле нуклеиновой кислоты. Т.к. G-C п. о. имеет 3, а А-Т — 2 водородные связи, то взаимодействие между нуклеотидами пары G-C более сильное, а соответственно и Тпл положительно коррелирует с долей пары G-C в молекуле ДНК.Англо-русский толковый словарь генетических терминов > melting temperature
